Effects of the Mixture of Eucommia ulmoides Oliver and Achyranthes japonica Nakai Extracts (KGC08EA) on Testosterone Synthesis in Late-Onset Hypogonadism (LOH) in vitro Models

Abstract

|
Late-onset hypogonadism (LOH) is associated with a significant decrease in the testosterone levels resulting from the reduced ability to synthesize testosterone in the Leydig cells as aging progresses. This study measured the testosterone levels after treating H2O2-stressed TM3 cells in the presence of human chorionic gonadotropin (hCG) with Eucommia ulmoides Oliver extracts (eu), Achyranthes japonica Nakai extracts (aj), or amixture of eu and aj extracts in the proportion of KGC08EA (1:3, 1:1, 3:1). KGC08EA (3:1) at concentrations of 1 to 500 ¥ìg/mL produced a significant concentration-dependent increase (up to 34 times (P<0.001)) in testosterone levels compared to the control group. In addition, among the testosterone synthesis enzymes, 17¥â-HSD1 and 3¥â-HSD4 showed significant increases in mRNA expression while both 5¥á-reductase2 and aromatase, such as testosterone degradation enzyme, was not increased significantly (P<0.05). In conclusion, KGC08EA (3:1) might bea health functional food material that can improve LOH by up-regulating the testosterone synthesis enzymes and increasing the testosterone levels significantly.
